چکیده

Background. The purpose of this study was to evaluate the effects of ASCs on full-thickness skin grafts. Specifically, we investigated the anti-inflammatory effects of ASCs that are mediated via regulation of the phenotypes of activated macrophages. Methods. ASCs were isolated, cultured, and injected under full-thickness skin grafts in 15 rats (ASC group). An additional 15 rats served as controls (PBS group). Skin graft survival assessment and vascularization detection were assessed with H&E staining and laser Doppler blood flowmetry (LDF). The effects of ASCs on angiogenesis, anti-inflammation, collagen accumulation-promoting, and antiscarring were assessed. Results. We found that the skin graft survival rate was significantly increased in the ASC group. The neovascularization, collagen deposition, collagen type I to type III ratio, and levels of VEGF and TGF-β3 in the ASC group were markedly higher than those in the PBS group at day 14. Additionally, in the ASC group, the levels of iNOS, IL-1β, and TNF-α were remarkably decreased, whereas the levels of IL-10 and Arg-1 were substantially increased. Conclusions. Our results confirm that ASCs transplantation can effectively improve full-thickness skin graft survival. Additionally, the anti-inflammatory role of ASCs may indirectly contribute to skin graft survival via its effect on macrophage polarization.

Skin wound healing following the spray of human abdominal adipose tissue-derived mesenchymal stem cells in diabetic male rat

Introduction: Diabetes is rising worldwide and impaired wound healing is one of its major complications. This study aimed to determine the effects of adipose-derived mesenchymal stem cells (MSCs) on wound healing in diabetic rats. Materials and Methods: In this experimental study, abdominal adipose tissue was obtained from 10 patients who underwent an abdominoplasty. MSCs were isolated from adi...

Healing Potential of Mesenchymal Stem Cells Cultured on a Collagen-Based Scaffold for Skin Regeneration

Background: Wound healing of burned skin remains a major goal in public health. Previous reports showed that the bone marrow stem cells were potent in keratinization and vascularization of full thickness skin wounds. Methods: In this study, mesenchymal stem cells were derived from rat adipose tissues and characterized by flowcytometry. Staining methods were used to evaluate their differentiatio...

تاثیر سلول‌های بنیادی برگرفته از چربی بر افزایش بقای گرافت غضروفی خرد شده در خرگوش

Background: In recent years the use of diced cartilage grafts in reconstructive surgery particulary rhinoplasty have been considered by most plastic surgeons. However, long-term resorption usually occurs. Stem cells are a powerful tool for reconstructive surgery to rebuild and maintain tissue with reduced complications. Since the adipose tissue-derived stem cells (ADSCs) can rebuild a wide vari...

Evaluation of the Effects of Adipose Derived Mesenchymal Stem Cells Cultured on Decellularized Amniotic Membrane in Wound Healing of Distal Part of the Limbs in Horse

Background: Equine wounds, particularly those involving the distal portion of the limbs, often undergo prolonged complex healing and may enter a non-healing state with obvious financial and welfare implications. Poor blood supply and lower levels of oxygen to the lower limbs lead to imbalance of growth factors related to wound healing. Decellular amniotic membrane has been widely used in soft t...
